The invention utilizes virtual screening strategy to seek for current market drugs as anti-schizophrenia therapy drug repurposing. Drug repurposing strategy finds new uses other than the original medical indications of existing drugs. Finding new indications for such drugs will benefit patients who are in needs for a potential new therapy sooner since known drugs are usually with acceptable safety and pharmacokinetic profiles. In this study, repurposing marketed drugs for DAAO inhibitor as new schizophrenia therapy was performed with virtual screening on marketed drugs and its metabolites. The identified and available drugs and compounds were further confirmed with in vitro DAAO enzymatic inhibitory assay.
L’invention utilise une stratégie de criblage virtuel pour rechercher des médicaments courants du marché pour leur réorientation en tant que médicament de thérapie contre la schizophrénie. La stratégie de réorientation de médicaments trouve de nouvelles utilisations autres que les indications médicales initiales de médicaments existants. Trouver de nouvelles indications pour de tels médicaments sera bénéfique pour des patients qui ont besoin d’une nouvelle thérapie potentielle plus rapidement puisque les médicaments connus sont habituellement associés à des profils pharmacocinétiques et d’innocuité acceptables. Dans cette étude, la réorientation de médicaments commercialisés en tant qu’inhibiteurs de DAAO en tant que nouvelle thérapie contre la schizophrénie a été effectuée avec un criblage virtuel sur des médicaments commercialisés et leurs métabolites. Les médicaments et composés identifiés et disponibles ont été en outre confirmés avec un essai d’inhibition enzymatique de DAAO in vitro.
