實驗先以28株益生菌進行體外抗氧化能力之測試,從中發現Lb. kefiranofaciens HL1這株菌較其他菌株有顯著較高的清除1,1-二苯基-2-三硝基苯肼(1, 1-diphenyl-2-picrylhydrazyl, DPPH)自由基及螯合鐵離子能力,並且可顯著抑制亞麻油酸過氧化,因此便以此菌株餵予以D-galactose誘導老化之小鼠,並對小鼠進行Morris水迷宮試驗之行為觀察及腦部海馬迴cleaved caspase-3表現量,以評估小鼠學習及記憶能力與海馬迴受損程度。實驗結果發現,攝取挑選之益生菌可顯著增強小鼠長期、短期及空間上的學習與記憶能力,並且餵予益生菌可顯著減少小鼠海馬迴中的cleaved caspase-3的表現量(P 0.05),顯示特定益生菌之攝取可以減緩老化造成的行為退化現象。再來,進一步透過小鼠體內抗氧化酵素、MDA濃度及菌相之分析,探討益生菌延緩老化可能機制之實驗結果中,發現餵予老化小鼠Lb. kefiranofaciens HL1可降低小鼠肝臟及腦部MDA濃度;另外,所有益生菌組皆表現較高的血液超氧歧化酶(superoxide dismutase, SOD)及過氧化氫酶(catalase)活性,並且其中高劑量HL1組腦部SOD活性及肝臟catalase活性顯著高於負對照組(P 0.05)。而在腸道微生物分析的結果可以發現益生菌組有較高乳酸桿菌屬(Lactobacillus),較低腸桿菌科(Enterobacteriaceae)比例之現象。綜合以上結果得到,利用體外抗氧化之測定可作為先前篩選菌株的方式,並且由結果推測益生菌帶來之延緩老化現象,可能是透過增強體內抗氧化的機制。
接著以Lb. kefiranofaciens HL1作為發酵菌元,開發出一種新穎的發酵乳製品,並以D-galactose誘導BALB/c小鼠老化模式評估其抗氧化能力、抗發炎和抗衰老之功效,推論其可能的途徑及腸腦軸之間的關聯。結果再度證明給予HL1單菌株之菌液能顯著的改善小鼠之學習力及長期空間記憶能力,顯著降低血清中TNF-α及IL-1β、肝臟中TNF-α、大腦中TNF-α及MDA的濃度,並顯著增加大腦中SOD的濃度。而給予FM共同發酵乳則對於改善小鼠之短期空間記憶能力有較顯著的效果。在分子層級上, 發酵乳組能夠顯著降低血清中IL-1β、肝臟中TNF-α、大腦中TNF-α及MDA的濃度,並顯著增加大腦中SOD以及在盲腸內容物中所含的丁酸濃度。最後,我們使用qPCR分析了盲腸微生物菌相組成的變化。與NC組相比,FM組能顯著降低產氣莢膜梭菌的比例。

受益於疫苗和抗生素發展、手術的改善與對疾病更多的研究及更佳的治療,人類壽命得已延長。臺灣行政院主計總處統計,2022年底,65歲以上人口占總人口17.56%,共計超過400萬人,國家發展委員會預估,2025年,老年人口比例將突破20%,即進入超高齡社會。人口高齡化是全球未來的趨勢,老化會使人體失去活動力、反應力及正常生理機能,導致許多年齡相關之慢性疾病,如:肥胖症、高血壓、糖尿病、心血管疾病、慢性腎臟病等,使病人喪失日常生活機能,也造成親屬照顧上的身心理壓力,因此如何維持健康、預防老化及老年疾病至關重要。
According to statistics from the World Health Organization (WHO) in 2022, the global population aged 60 and above exceeded the population under 5 years old in 2020. WHO also estimates that the population aged 60 and above will nearly double from 2015 to 2050, rising from 12% to 22%. In other words, by 2050, the global population aged 60 and above will reach a staggering 2.1 billion. According to the National Statistics Report of the Directorate-General of Budget, Accounting and Statistics, by the end of February 2017, Taiwans aging index surpassed 100 for the first time, indicating that the population aged 65 and above has exceeded the population under 14. According to statistics from the Ministry of the Interior, as of the end of January 2021, Taiwans population aged 65 and above was 3.804 million, accounting for 16.2% of the total population. With the global trend of population aging, national health and social systems face daunting challenges. The extension of the elderly stage should be built on a foundation of health. Therefore, disease prevention and care for the elderly are crucial. Enhancing the quality of life for caregivers will be a future market trend.

目前研究發現老化、慢性疾病及腸道菌相改變之關連性。腸道菌相失衡可能破壞腸道屏障,使腸道通透性增加及誘發腸道免疫反應,進而引起慢性發炎並導致許多慢性疾病,在某些情況下,血腦屏障被破壞,引起神經發炎、神經損傷和神經元損失,最終導致神經退化性疾病。已知某些特定益生菌株具有抗氧化、抗發炎和延緩衰老之功效,可改善腸道菌相失衡、降低氧化壓力及全身性慢性發炎,或藉由腸腦軸減緩神經損傷及神經退化性疾病。目前沒有抗老化特效藥,藥品不僅需要處方箋,也都有一定的副作用,因此益生菌為老化所造成之身體機能衰退及心理症狀提供一種更天然且健康的預防性措施。
